Smoothened Agonist (SAG) HCl是一種細胞滲透性Smoothened (Smo)激動劑，其在Shh-LIGHT2細胞中的EC50約為3 nM

靶點：

Smoothened(mouse cultured cell assay):3 nM(EC50);Hedgehog/Smoothened

體外研究：

SAG通過直接結合到Smo螺旋束調(diào)控Smo活性。 SAG以GRK2依賴的方式誘導通過Gli的Smo-依賴性信號。 SAG(1 nM)也會誘導神經(jīng)元和神經(jīng)膠質(zhì)前體細胞的增殖，而不影響新生細胞的分化模式

體內(nèi)研究：

在成年大鼠海馬中，SAG (2.5 nM)腦室內(nèi)給藥顯著增加新生細胞的數(shù)量，并延長海馬細胞的存活時間。在小鼠中，SAG (20 μg/g, i.p.)有效防止GC-誘導的新生小腦發(fā)育異常

細胞實驗：

Cell lines: Shh-LIGHT2 cells Concentrations: 3 nM Incubation Time: 30 h Method: Cells were treated with various concentrations of SAG in DMEM containing 0.5% (vol/vol) bovine calf serum.

動物實驗：
